ABSTRACT

Angelicae Sinensis Radix (AS) is reproted to exert anti-depression effect (ADE) and nourishing blood effect (NBE) in a rat model of depression. The correlation between the two therapeutic effects and its underlying mechanisms deserves further study. The current study is designed to explore the underlying mechanisms of correlation between the ADE and NBE of AS based on hepatic metabonomics, network pharmacology and molecular docking. According to metabolomics analysis, 30 metabolites involved in 11 metabolic pathways were identified as the potential metabolites for depression. Furthermore, principal component analysis and correlation analysis showed that glutathione, sphinganine, and ornithine were related to pharmacodynamics indicators including behavioral indicators and hematological indicators, indicating that metabolic pathways such as sphingolipid metabolism were involved in the ADE and NBE of AS. Then, a target-pathway network of depression and blood deficiency syndrome was constructed by network pharmacology analysis, where a total of 107 pathways were collected. Moreover, 37 active components obtained from Ultra Performance Liquid Chromatography-Triple-Time of Flight Mass Spectrometer (UPLC-Triple-TOF/MS) in AS extract that passed the filtering criteria were used for network pharmacology, where 46 targets were associated with the ADE and NBE of AS. Pathway enrichment analysis further indicated the involvement of sphingolipid metabolism in the ADE and NBE of AS. Molecular docking analysis indciated that E-ligustilide in AS extract exhibited strong binding activity with target proteins (PIK3CA and PIK3CD) in sphingolipid metabolism. Further analysis by Western blot verified that AS regulated the expression of PIK3CA and PIK3CD on sphingolipid metabolism. Our results demonstrated that sphingolipid metabolic pathway was the core mechanism of the correlation between the ADE and NBE of AS.

ABSTRACT

Objective:To explore the application value of modified Shiquan Dabutang in the treatment of elderly patients with osteoporotic intertrochanteric fractures (OIFs) due to Qi and blood deficiency by observing its impacts on inflammatory and bone metabolism indexes. Method:Ninety-eight elderly patients admitted to our hospital for OIFs of Qi and blood deficiency syndrome from April 2018 to April 2020 were randomized into an observation group (<italic>n</italic>=49) and a control group (<italic>n</italic>=49). Following the proximal femoral nail antirotation (PFNA) fixation, patients in the control group were treated with Guipiwan, while those in the observation group received the modified Shiquan Dabutang. The clinical efficacy, inflammatory and bone metabolism indexes, and complications were compared between the two groups after four weeks of treatment. Result:The levels of such serum indexes as fibroblast growth factor-2 (FGF-2), osteoprotegerin (OPG), transforming growth factor-<italic>β</italic><sub>1</sub> (TGF-<italic>β</italic><sub>1</sub>), <italic>β</italic>-endorphin (<italic>β</italic>-EP), bone-specific alkaline phosphatase (BALP), and osteocalcin (BGP) in the observation group after treatment were significantly elevated as compared with those in the control group (<italic>P</italic><0.05), whereas the serum tumor necrosis factor-<italic>α</italic> (TNF-<italic>α</italic>) and <italic>D</italic>-dimer (<italic>D</italic>-D) declined (<italic>P</italic><0.05). The TCM symptom score in the observation group after treatment was obviously lower than that in the control group, while the Harris Hip Score (HHS) was higher (<italic>P</italic><0.05). The overall response rate of the observation group was 93.88% (46/49), higher than 75.51% (37/49) of the control group (<italic>χ<sup>2</sup></italic>=6.376, <italic>P</italic><0.05). The total incidence of incision infection, deep vein thrombosis of lower limbs, and pulmonary infection in the control group was 24.49% (12/49), significantly higher than 6.12% (3/49) in the observation group (<italic>χ<sup>2</sup></italic>=6.607, <italic>P</italic><0.05). Conclusion:The modified Shiquan Dabutang is able to alleviate inflammation, regulate bone metabolism, promote bone repair, and reduce the incidence of complications in elderly patients with OIFs due to Qi and blood deficiency.

ABSTRACT

Objective:To observe the effect of modified Bazhentang on the nutritional status and immune function of patients with Qi and blood deficiency syndrome in neoadjuvant chemotherapy (NAC) for gastric cancer. Method:One hundred and ten patients were randomly divided into observation group and control group with 55 cases each. Both groups accepted FOLFOX6 protocol. Patients in control group took Jianpi Shengxue tablets orally, 3 tablets/time, 3 times/day. Patients in observation group received modified Bazhentang, 1 dose/day. The course of treatment was six weeks in both groups. Before and after treatment, scores were graded according to patient generated-subjective global assessment (PG-SGA), Qi and blood deficiency syndrome, and the Revised Piper Fatigue Scale (PFS-R). Levels of serum total protein (TB), albumin (ALB), prealbumin (PAB), CD4<sup>+</sup>, CD8<sup>+</sup>, helper T lymphocyte 17 (Th17), regulatory T cell (Treg), immunoglobulin G (IgG), IgM, and IgA were detected before and after therapy. Body mass index (BMI) and fat free mass index (FFMI) were measured before and after treatment. Weight loss was recorded, and the acute or subacute toxicity of anticancer drugs was evaluated. Result:The degree of malnutrition in the observation group was lower than that in the control group (<italic>Z</italic>=2.401,<italic>P</italic><0.01). The levels of TB, ALB and PAB in the observation group were higher than those in the control group (<italic>P</italic><0.01). The CD4<sup>+</sup>, Treg and CD4<sup>+</sup>/CD8<sup>+</sup> levels in the observation group were higher than those in the control group (<italic>P</italic><0.01). The CD8<sup>+</sup>, Th17 and Th17/Treg levels were lower than those in the control group (<italic>P</italic><0.01). Besides, the levels of IgM and IgA in the observation group were higher than those in the control group (<italic>P</italic><0.01). The PG-SGA score and weight loss in the observation group were lower than those in the control group (<italic>P</italic><0.01). The BMI and FFMI data of the observation group were higher than those of the control group (<italic>P</italic><0.05). The scores of PFS-R and Qi-blood deficiency syndrome were lower than those of the control group (<italic>P</italic><0.01). The incidence of nausea and vomiting in the observation group was 45.45% (25/55), lower than 65.45% (36/55) in the control group (<italic>χ</italic><sup>2</sup>=4.452,<italic>P</italic><0.05). Conclusion:Modified Bazhentang can be used to assist gastric cancer patients with NAC, which can improve nutritional status and immune function, promote immune balance, reduce clinical symptoms and fatigue, and reduce chemotherapy toxicity and side effects, so it is worthy of clinical use.

ABSTRACT

Objective:To observe the effect of Dingkundan in adjuvant treatment of clinical symptoms, quality of life, immune function and prognosis of patients with radiotherapy and chemotherapy after endometrial carcinoma (EC) operation. Method:Patients were divided into control group (82 cases) and observation group (86 cases) according to random number table. A total of 75 patients in control group completed the study (4 patients fell off or lose visit, and 3 patients were eliminated), while 77 patients in observation group completed the study (5 patients fell off or lose visit, and 4 patients were deleted). After operation, patients got brachytherapy, external pelvic irradiation and chemotherapy. Patients in control group got Bazhenwan, 1 pill/time, 2 times/day, and those in observation group got Dingkundan, 7 g/time, 2 times/day. The course of treatment lasted for 4 months, and long-time follow-up data was recorded. Before treatment, and at the second and fourth month after treatment, deficiency of Qi and blood was scored. Toxic reactions after radiotherapy and chemotherapy were recorded, and incidence rate of acute and advanced radiation injury of bladder and rectum and toxicity of chemotherapeutic drugs at grade 3 or above grade 3 level were compared. And levels of T lymphocyte subsets (CD3<sup>+</sup>, CD4<sup>+</sup>, CD8<sup>+</sup> and CD4<sup>+</sup>/CD8<sup>+</sup>) were detected, European collaborative quality of life Cancer Core Scale (EORTC QLQ-C30) was evaluated, and expressions of pce125 (CA125), epididymis protein 4 (HE4), Dickkopf-related protein-1 (DKK1), vascular endothelial growth factor (VEGF), matrix metalloproteinase-9 (MMP-9) and transforming growth factor-<italic>β</italic><sub>1</sub> (TGF-<italic>β</italic><sub>1</sub>) were tested before and after treatment. The follow-up was made for every three months, and the progression (recurrence/metastasis) of patients was recorded. Result:Scores of deficiency of Qi and blood in observation group were lower than those in control group at the second and fourth week after treatment (<italic>P</italic><0.01). Incidence rates of acute and advanced radiation injury of bladder and rectum and toxicity of chemotherapeutic drugs at grade 3 or above grade 3 level and incidence rates of bone marrow suppression, gastrointestinal toxicity, neurotoxicity were lower than those in control group (<italic>P</italic><0.05). Five functional dimensions and overall quality of life score based on EORTC and QLQ-C30 in observation group were higher than those in control group (<italic>P</italic><0.01), and scores of three symptom dimensions were lower than those in control group (<italic>P</italic><0.01). Levels of CD3<sup>+</sup>, CD4<sup>+</sup> and CD4<sup>+</sup>/CD8<sup>+</sup> were higher than those in the control group (<italic>P</italic><0.01), and CD8<sup>+</sup> was lower than those in the control group (<italic>P</italic><0.01). Levels of CA125, HE4, DKK1, VEGF, MMP-9 and TGF-<italic>β</italic><sub>1</sub> were lower than those in the control group (<italic>P</italic><0.01). The disease progression rate in observation group was 18.18% (14/77), which was lower than 33.33% (25/75) in control group (<italic>χ</italic><sup>2</sup>=4.572, <italic>P</italic><0.05). Conclusion:In adjuvant treatment of patients with radiotherapy and chemotherapy after EC operation, Dingkundan can reduce the symptoms of Qi and blood deficiency syndrome and side effects caused by radiotherapy and chemotherapy, improve the quality of life and immune function, inhibit the expression of tumor markers and tumor growth factor, delay the progression of tumor and improve the prognosis.

ABSTRACT

Objective:To study the clinical efficacy of modified Shengyutang on patients with active stage psoriasis vulgaris due to Qi and blood deficiency. Method:The 134 cases were randomly divided into control group and observation group, with 67 cases in each group. The control group was given avic a capsule + Danggui Buxuewan, while the observation group was given avic a capsule + modified Shengyutang for 4 weeks, respectively. The psoriasis area and severity index (PASI), dermatological life quality index (DLQI) and psoriasis vulgaris due to Qi and blood deficiency syndrome were observed before and after treatment. The serum growth factor [endothelial cell specific molecule-1 (ESM-1), transforming growth factor-<italic>β</italic><sub>1</sub>(TGF-<italic>β</italic><sub>1</sub>), vascular endothelial cell growth factor (VEGF)], hemorheological indicators [high cut blood viscosity (HBV), low cut blood viscosity (LBV), erythrocyte sedimentation rate (ESR)], CC cphenotype receptor(CCR)6, CC cphenotype ligand 20 (CCL20), monocyte chemotactic protein-4 (MCP-4) in serum and tissue fluid of lesions were detected. Clinical efficacy and recurrence follow-up for 12 months were compared. The safety was evaluated between two groups. Result:Three cases in control group and one case in observation group fell off during the study period. The total effective rate was 96.97% (64/66) in observation group, which was higher than 81.25% (52/64) in control group (<italic>χ</italic><sup>2</sup>=5.064, <italic>P</italic><0.05). During the 12-month follow-up, the recurrence rate was 20.31% (13/64) in observation group, which was lower than 51.92% (27/52) in control group (<italic>χ</italic><sup>2</sup>=6.038, <italic>P</italic><0.05). Compared with control group after treatment, PASI, DLQI, TCM syndromes, ESM-1, TGF-<italic>β</italic><sub>1</sub>, VEGF, HBV, LBV, ESR, CCR6, CCL20 and MCP-4 in observation group were significantly reduced (<italic>P</italic><0.05). No obvious blood and urine routine, or heart, liver and renal dysfunction was observed in the two groups. The incidence of adverse reactions was 3.03% (2/66) in observation group, which was lower than 26.56% (17/64) in control group (<italic>χ</italic><sup>2</sup>=5.764, <italic>P</italic><0.05). Conclusion:Modified Shengyutang can significantly improve the clinical symptoms of patients with active stage psoriasis vulgaris due to Qi and blood deficiency, with a low recurrence rate and the incidence of adverse reactions.

ABSTRACT

Arthritis of the carpometacarpal joint of the thumb is typically treated with oral anti-inflammatory drugs, external medicines, and temporary external fixation. We report 3 cases of arthritis of carpometacarpal joint of the thumb that were successfully treated with sokeikakketsuto. In cases 1 and 2, a 50-year-old woman and 68-year-old man, respectively, were diagnosed with arthritis of the carpometacarpal joint and were successfully treated with sokeikakketsuto. In case 3, a 66-year-old man complained of pain in the second carpometacarpal joint caused by gout, and was successfully treated with eppikajutsuto and daiobotampito. However, the pain in the carpometacarpal joint of the thumb due to arthritis remained, and was successfully treated with sokeikakketsuto. Eppikajutsuto and daiobotampito were effective for acute arthritis such as gout, and sokeikakketsuto was effective for osteoarthritis. Overall, patients with different diseases in the various local regions may have slightly different clinical presentations even in the same case.

@#AIM: To evaluate effects of Qing Mang Yi Hao in the treatment of toxic optic atrophy.<p>METHODS: A retrospective analysis of 7 patients(13 eyes)suffered from toxic optic atrophy in Department of Ophthalmology, Dongfang Hospital Beijing University of Chinese Medicine, who were diagnosed as liver stagnation and blood deficiency and treated with Qing Mang Yi Hao formula. The formula treatments patients received were varying from 1 to 12 courses, and the clinical follow-up unequally lasted from 6-24mo. Then the formula clinical efficacy was comprehensively evaluated by comparing their visual acuity, visual field, visual evoked potential(VEP)and retinal nerve fiber layer(RNFL)thickness before and after treatment.<p>RESULTS: Seven cases(13 eyes)consisted of 5 ethambutol-induced optic neuropathy cases(10 eyes), 1 alcohol toxic neuropathy case(1 eye)and 1 rabies vaccine toxic neuropathy case(2 eyes). OCT showed the RNFL thicknesses on nasal and temporal quadrants were significantly thin at their first visits and the tendency of decreasing was noticed on each quadrant of RNFL thickness at the last follow-up. However, the visual acuity of 7 cases(13 eyes)had varying improvements: ≥0.1 4 eyes(31%), 0.06-<0.1 2 eyes(15%), 0.04-<0.06 1 eyes(8%), 0.01-<0.04 4 eyes(31%), no improvements 2 eyes(15%). The total efficacy rate of Qing Mang Yi Hao formula was 64%(27% significant).<p>CONCLUSION: Qing Mang Yi Hao could improve visual function, delay the progressive optic atrophy of TON, thus plays a significant role in optic neuroprotection.

ABSTRACT

Objective::To explore the clinical efficacy of Magui Wenjingtang combined with joint mobilization in the treatment of periarthritis of the shoulder and the effect on hemodynamics and serum pain media. Method::A total of 120 patients with scapulohumeral periarthritis were enrolled in Tianjin Ninghe Hospital from July 2016 to July 2018.All cases were divided into control group and observation group, with 60 patients in each group. The control group received shoulder joint loosening operation (qd), and celecoxib capsules for oral administration. The observation group was treated with Magui Wenjingtang in addition to the shoulder joint loosening operation, 1 dose/d, 2 times/d. After 4 weeks of continuous treatment, the subjects were compared for the subjective pain level McGill Pain Questionnaire (SF-MPQ) score, shoulder mobility hemodynamic changes, and serum prostaglandin E2 (PGE2), substance P (SP), serotonin (5-HT) level and clinical efficacy. Result::Compared with before treatment, visual pain score (VAS), current pain status (PPI), and pain rating index (PRI) scores of the two groups were significantly lower after 4 weeks of treatment, and those of the observation group was significantly lower than the control group (P<0.01). The range of shoulder joint activity was significantly higher than that of the control group (P<0.01), erythrocyte sedimentation rate (ESR), fibrinogen (Fb), whole blood viscosity and serum were compared between the two groups after 4 weeks of treatment and before treatment. The levels of PGE2, SP and 5-HT were significantly lower (P<0.01), and the observation group was significantly better than the control group (P<0.01). The total effective rate of the observation group was 96.61%(57/59), which was significantly higher than 82.76% (48/58) of the control group(χ2=4.685, P<0.05). Conclusion::Magui Wenjingtang combined with shoulder joint loosening operation in treating periarthritis of shoulder with syndrome of blood deficiency and cold coagulation can effectively improve the pain and shoulder joint mobility, increase the curative effect, and improve the hemodynamics, and down-regulatie pain mediator of PGE2, SP, 5-HT, which may be related to the curative effect.

ABSTRACT

Six patients with psychiatric symptoms were successfully treated with shimotsuto-containing medication. Case 1 was a 13-year-old boy who complained of difficulty in getting up ; however, when he was treated with a focus on the decrease in his concentration, he was able to go to school after treatment. Case 2 was an 18-year-old girl who had also difficulty in getting up, but was able to go to school. Case 3 was a 10-year-old girl who was able to go to school partially after treatment. Case 4, a 42-year-old man with apprehension in socializing, showed improvement in socializing after taking medication containing shimotsuto. Case 5 had various complaints that were relieved after taking medication containing shimotsuto. Case 6 was a 56-year-old woman who had also multiple complaints ; however, the symptoms were alleviated with shimotsuto-containing medication. All patients experienced depression and anxiety. Four patients with decreased cortisol recovered after treatment with shimotsuto. From these results, shimotsuto is considered to improve concentration, judgment, and psychiatric symptoms by treating blood deficiency.

ABSTRACT

The main symptoms of fibromyalgia are systemic pain and fatigue. The pharmacological treatments are usually performed for fibromyalgia in Western medicine. However, in some cases, pain that impairs everyday life continues even after medical treatment. We herein report a case in which fibromyalgia was successfully treated using jumizasan. The patient was a 28-year-old woman. Prior to admission to the author's clinic, she had complained of dull pain in the left upper arm for 4 years, and the pain had spread throughout the body for 2 years. Various other symptoms including fatigue, headache, heart palpitation, and shortness of breath coexisted. The pain resolved within 3 months after the initiation of treatment with jumizasan (a Japanese herbal medicine decoction), which was given based on pallid complexion, skin drying, strong upper arm pain, and walking difficulty. We concluded that jumizasan may be considered to be used for patients with blood deficiency, upper arm pain, and walking difficulty in Kampo concept of treatment.

Objective: Through metabonomics research methods,the effect of Siwutang on metabolites and metabolic pathways in natural aging mice were observed.The related targets and mechanism of Siwutang intervention in natural aging mice were analyzed. Method: Taking 20-month-old natural aging model mice(equivalent to 60-65 years old of human beings) as the experimental subjects,at the same time,mice aged 3 months were established as the youth group.UPLC-Q-TOF-MS technique was employed to analyze the mouse plasma with mobile phase of acetonitrile(containing 0.1%formic acid)-0.1%formic acid solution for gradient elution and positive ion mode of electrospray ionization,and the metabolic markers were analyzed by principal component analysis(PCA) and orthogonal partial least squares-discriminant analysis(OPLS-DA),and their metabolic pathways were summarized. Result: Siwutang had obvious reversal effect on the expression levels of 16 aging-related metabolites,among which 9 metabolic markers were statistically significant(PConclusion: Siwutang can affect the metabolites in the plasma of 20-month-old natural aging mice,and the metabolic disorder during the aging process of mice can be improved by glutathione metabolism,pyrimidine metabolism,selenium amino acid metabolism and other pathways,and this paper can provide biological information for the study of material basis of this compound for aging.

ABSTRACT

In this paper,ultra performance liquid chromatography coupled with time-of-flight mass spectrometry( UPLC-Q-TOFMS) technique was used to study the effects of steamed notoginseng on endogenous markers in plasma of rats with hemolytic anemia induced by N-acetyl phenyl hydrazine( APH). The aim was to find out the potential biomarkers and possible blood enriching mechanism of steamed notoginseng on hemolytic anemia rats. In the experiment,steamed notoginseng medicine pair( steamed notoginseng-ginseng)and compound medicines( Sanqi Yangxue Capsules) were used respectively to intervene in APH-induced hemolytic anemia model rats.Then blood routine indexes such as red blood cells( RBC),hemoglobin( Hb) and related organ indexes were determined. As compared with the blank group,the RBC and Hb levels in the model group were substantially decreased( P< 0. 01),while the liver and spleen organ indexes were increased( P< 0. 05). The results of blood routine and organ index demonstrated that the blood deficiency model was successfully established. Steamed notoginseng can significantly increase the RBC level of rats( P<0. 01),and the related indicators of each drug group had a trend of returning to normal levels,verifying the blood enriching effect of steamed notoginseng. The UPLC-Q-TOF-MS technique,principal component analysis( PCA) and partial least squares-discrimination analysis( PLS-DA) were used to analyze the metabolic profiles between the normal group and the model group. Twenty-six potential biomarkers for hemolytic anemia were screened in plasma. Nine metabolites such as retinol,L-valine,and arachidonic acid were down-regulated in the blood deficiency rats,and 17 metabolites such as protoporphyrin Ⅸ and niacinamide were up-regulated. The metabolic level of biomarkers could be changed to a normal state after rats were given with steamed notoginseng,drug pairs,and compound prescriptions. It can be speculated that steamed notoginseng may play a role of blood tonifying by improving biosynthesis of valine,leucine and isoleucine,as well as metabolic pathways such as retinol metabolism and arachidonic acid metabolism.

ABSTRACT

Ultra performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(LC-MS) was used to establish the chromatography fingerprint for fresh(FRAS) and dry(RAS) roots of Angelica sinensis from 10 different places. The rat model of blood deficiency was established by acetyl-phenyl-hydrazine(APH) and cyclophosphamide(CTX). Then grey relational analysis(GRA) and partial least squares regression(PLS) were used to investigate the spectrum-effect relationship between the relative contents and the data of enriching blood pharmacodynamics efficacy. The results showed that the FRAS and RAS had certain enriching blood activities(P<0.05). The contribution degree of the FRAS and RAS to enriching blood activities of each common peaks were determined by regression coefficient. Among them, 4 common peaks contributed significantly to the effect of enriching blood activities, P1(unknown), P2(unknown), P7(ferulic acid), and P11(senkyunolide A) respectively. This paper investigated the spectrum-effect relationship between enriching blood activities and LC-MS chromatography fingerprint of RAS and FRAS, and determined the effective compositions of RAS and FRAS with enriching blood activities. It lays a theoretical foundation for the comprehensive development and utilization of A. sinensis.

ABSTRACT

Hypertensive urgency is sometimes repeated and difficult to treat with anti-hypertensive agents or anxiolytics. There were few reports on treatment of hypertensive urgency with Kampo medicine. We experienced five cases of frequent hypertensive urgency, successfully treated with shichimotsukokato only. All cases had deficiency pattern and symptoms of blood deficiency. It took relatively short term to stabilize blood pressure after taking shichimotsukokato, and the efficacy of this drug had been continued. Shichimotsukokato can attribute to rapid and continuous stabilization of blood pressure for frequent hypertensive urgency of those who have deficiency pattern and blood deficiency.

ABSTRACT

We evaluated body constituents patterns of 74 consecutive patients with neurological degenerative disorders. They comprise Parkinson's disease (n = 38), amyotrophic lateral sclerosis (n = 19), and multiple system atro­phy (n = 17). We compared body constituents patterns between them and 149 consecutive patients with other neurological diseases of the same age. We used ki-ketsu-sui scores to evaluate body constituents patterns in all cases. Ki-ketsu-sui scores measure six factors : qi deficiency (kikyo), qi stagnation (kiutsu), qi counterflow (kigyaku), blood deficiency (kekkyo), blood stasis (oketsu), and fluid retention (suitai). As a result of multi­variate analysis, neurological degenerative disorders had large weight of blood deficiency, fluid retention and qi stagnation. Their adjusted odds ratios (95% confidence interval) were 3.02 (1.43-­6.48), 2.37 (1.13-­5.11), 2.33 (1.01-­5.44), respectively. Most relevant factor to neurological degenerative disorders was a blood defi­ciency. Taking into consideration a prescription of “shimotsuto rui” may contribute to alleviate patient's suf­fering. In addition to subjective symptoms, we need an oriental medicine scale such as pulse, tongue, and ab­dominal examinations to judge a therapeutic effect of Kampo medicine.

ABSTRACT

Aim: To study the anti-fatigue,anti-oxidative and hemostatic effects of small molecule Asini Corii Colla (SMACC). Methods: Rat model of complex blood deficiency was established to detect the exhausted time of swimming, hematology, superoxide dismutase (SOD), serum maleic dialdehyde (MDA), and lipid peroxide (LPO) levels, aiming to explore the anti-fatigue and anti-oxidative effects of SMACC. ICR mice were used to measure the blood clotting time (CT) and bleeding time (BT). Fevered and bleeding model and the heparinized bleeding model in rats were established to investigate the effect of SMACC in hemostasis and its possible mechanism. Results: Compared with model group, SMACC significantly prolonged the swimming time of model rats (P < 0. 05, P < 0. 01) and decreased the content of serum MDA, LPO (P < 0.05,P<0.01) at doses of 1. 500,0. 750,0. 375 g. kg-1,and increased the number of lymphocytes in the blood at doses of 1. 500,0. 375 g. kg-1 (P <0. 05). SMACC of 3. 00,1.50 g kg-1 significantly reduced the BT and CT (P < 0. 05). SMACC markedly reversed the prolonged prothrombin time (P < 0. 05) and the adverse changes of the hematological indicators. Conclusions: SMACC has the pharmacological effects of anti-fatigue, anti-oxidation and enhancing endurance, and also the effects of hemostasis and convergence.

ABSTRACT

OBJECTIVE: To study the blood enriching effect and mechanism of Jiuweiejiao ointment on mouse model of blood deficiency syndrome.METHODS: Blood deficiency syndrome model was established with cyclophosphamide(CTX) and acetylphenylhydrazine(APH). Total of 60 ICR mice were divided into normal group, model group, Siwu granule group, Jiuweiejiao ointment high-dose, medium-dose and low-dose group (15.6, 7.8, 3.9 g•kg-1). Each group mouse were treated with the respective drug through intragastrical administration once a day for consecutive 14 d. The levels of red blood cells (RBC), leukocytes (WBC), hemoglobin (HGB) level in blood of mice were detected by blood analyzer; the histopathological changes of bone marrow were determined by HE staining; the levels of erythropoietin (EPO), interleukin-3 (IL-3), tumor necrosis factor-α (TNF-α), granulocyte colony stimulating factor (G-CSF) and granulocyte-macrophage colony stimulating factor (GM-CSF) in serum were measured by enzyme linked immunosorbent assay (ELISA) method; the expression level of EPO protein in kidney tissue, G-CSF and GM-CSF protein in bone marrow tissue were detected by Western-blot method.RESULTS: Compared with model group, Jiuweiejiao ointment could significantly increase the levels of RBC, WBC and HGB in blood of mice, markedly improve bone marrow hyperplasia, reduce the non-hematopoietic tissue and adipocytes, obviously repaire the hematopoietic tissue structure; significantly decrease the level of TNF-α and increase the level of EPO, IL-3, G-CSF and GM-CSF in serum. In addition, Jiuweiejiao ointment could also obviously increase the expression of EPO protein in kidney tissue and G-CSF, GM-CSF protein in bone marrow tissue compared with that in model group.CONCLUSION: Jiuweiejiao ointment has obvious blood enriching effects, which may be related with promoting the secretion of hematopoietic growth factors and reducing the inhibition of hematopoietic function by negative hematopoietic factor.

ABSTRACT

Objective To observe the effects of Angelica Sinensis Radix on the expression of aquaporin 8 (AQP8) and the levels of AC-cAMP-PKA in colon of blood-deficiency constipation in mice; To identify mechanism of Angelica Sinensis Radix for loosening the bowels to relieve constipation. Methods Sixty Kunming mice were randomly divided into control group, model group, positive medicine group, and Angelica Sinensis Radix high-, medium-, and low-dose groups, with ten mice in each group. Diphenoxylate, acetylphenlyhydrazine and cyclophosphamide were used to establish blood-deficiency constipation mice models. From the 14thday of the experiment, Angelica Sinensis Radix high-, medium-, and low-dose groups were given 16.7, 8.8, and 4.2 g/kg Angelica Sinensis Radix Decoction for gavage. Positive medicine group was given 5.0 g/kg Changtongshu Granules Liquid for gavage. Control group and model group were given equal volume of saline for gavage. The symptoms of blood deficiency constipation were observed and defecation time. Immunohistochemistry, Western blot and qRT-PCR were used to detect the expression of AQP8 protein and mRNA in colon, and the expression of AC-cAMP-PKA in colon was detected by ELISA. Results The mice in the model group developed blood deficiency constipation syndrome; the defecation time was significantly prolonged (P<0.01); the expression level of colonic AQP8 protein and mRNA, AC, cAMP and PKA significantly increased (P<0.05, P<0.01). Compared with model group, the defecation time was significantly shortened in Angelica Sinensis Radix high-, medium-, and low-dose groups; the expression of AQP8 protein and mRNA and the levels of AC, cAMP and PKA were significantly decreased (P<0.05, P<0.01). Conclusion The treatment of Angelica Sinensis Radix for blood-deficiency constipation may be related to adjusting the AC-cAMP-PKA signaling pathways and reducing the expression of AQP8 protein and mRNA.

ABSTRACT

Objective To observe the clinical efficacy of collateral-pricking bloodletting therapy plusXiao Feng San in treating chronic urticaria due to blood deficiency and wind dryness.Method A total of 150 patients with chronic urticaria due to blood deficiency and wind dryness were randomized into a treatment group and a control group, 75 cases each. The treatment group was intervened by collateral-pricking bloodletting plus oral administration ofXiao Feng San, while the control group was intervened by oral administration ofXiao Feng San alone. Before and after 4-week treatment, the serum indexes [immunoglobulin E (IgE), eosinophil count, C3, C4, interleukin (IL)-2, IL-4] of the two groups were observed, and the clinical efficacies were also compared.Result The total effective rate and markedly effective rate were respectively 100.0% and 86.7% in the treatment group versus 80.0% and 52.0% in the control group, and the between-group differences were statistically significant (P<0.05,P<0.01). After the treatment, the serum indexes were significantly changed in the treatment group (P<0.05). The levels of IgE, eosinophil count, IL-2 and IL-4 were significantly changed after the treatment in the control group (P<0.05). There were significant differences in comparing the serum indexes between the two groups after the intervention (P<0.05).Conclusion Collateral-pricking bloodletting therapy plusXiao Feng San is an effective method in treating chronic urticarial due to blood deficiency and wind dryness, and it can improve the serum indexes.

ABSTRACT

Gastroesophageal reflux disease (GERD) can cause not only esophageal symptoms, but also extraesophageal symptoms such as globus pharyngis. Here, we describe a case of globus pharyngis in GERD successfully treated with seinetsuhoketsuto. A 72-year-old female began to have tingling in her pharyngolarynx in February 20XX. Otolaryngologist did not point out abnormal findings. However, reflux esophagitis was detected by upper gastrointestinal endoscopy. Although treatment with rabeprazole relieved the tingling, she experienced a relapse in September 20XX and visited our clinic in November. We prescribed seinetsuhoketsuto because she exhibited numerous blood deficiency-related symptoms including dryness of the skin and eyes, shallow sleep, coldness in the feet, etc. The tingling then decreased rapidly and disappeared in two months. In addition, dryness of the skin and eyes and her sleep also improved markedly. Seinetsuhoketsuto has been regarded as a medication for inflammation in the mouth such as oral ulcers and erosions. However, this case indicated that seinetsuhoketsuto could also be utilized to treat symptoms in the pharyngolarynx due to GERD.
